To identify a character other than the protagonist, you must examine the context clues of the story - little hints throughout the book that you can use to come to a conclusion. Pretend that you are like a detective gathering evidence as you read. For example, if you were to look for the story's antagonist (also known as the villain), you might search for signs that one of the characters is evil or detrimental to the protagonist (or hero) in some way. Antagonists tend to serve a role of fighting the hero of the story, so keep an eye out for any character who doesn't want your protagonist to succeed with their goal.
