Both prepositions are acceptable. However, assuming that everyone has some sort of accent, you would say they speak with that accent instead of in but if someone pretended to speak in a different accent, you would use in and not with.
I wish to speak WITH a British accent' or 'I wish to speak IN a British accent' - is there a difference?
