In both animal and plant cells, genomic DNA resides in the nucleus, which is surrounded by the nuclear membrane. Additionally, both types of cells are bound by a cell membrane. Plant cells, however, are enclosed by an additional cell wall composed of cellulose. There are two barriers in animal cells (i.e., nuclear membrane, cell membrane) and three barriers in plant cells (i.e., nuclear membrane, cell membrane, cell wall) that must be broken in order to extract genomic DNA.
