Graduated cylinders are used for measuring liquids. They're glass containers with measure marks on them (called graduations) kind of similar to a measuring cup but in a different shape.
Test tubes are used a lot of time to prepare small cultures or small amounts of a solution. I use plastic ones with a cap on it to store bacterial media so it can grow to a high enough density for the experiments I want. You may also see people use re-usable glass test tubes, which is especially useful when conditions don't need to be sterile.
Spot plates are ceramic or plastic well plates. They kind of look like a rectangular paint palette. You would use these for multiple small biological or chemical tests, like serial dilutions, comparative growth studies, or screening for viability with multiple strains or stressors. They're good for looking at quick visual changes for multiple things, especially changes in color or cloudiness.
