What is the difference between the verb Ser and Estar in Spanish?

Without getting too technical, both verbs mean "to be". However, SER is used with a long-lasting or permanent attribute whereas ESTAR is used with a shorter-term or variable characteristic.
For example, you would say "Ella ES inteligente". ES comes from the verb SER and intelligence is an intrinsic, long-lasting trait that fits the criteria for SER (i.e. a girl that is intelligent today will likely be intelligent in two years).
On the other hand, you would say "Ella ESTÁ enojada". ESTÁ comes from the verb ESTAR and means "angry" and since anger is a temporary state of emotion if would fit the criteria for ESTAR (i.e. a gril that is angry now may be perfectly calm in three hours).
There are obviously more nuances and exceptions but if you can understand the basic principle stated above it will give you a very strong foundation for understanding the difference between SER and ESTAR.
The easiest rule is: "How you feel and where you are always use the verb ESTAR", for anything else use SER.

The verbs "Ser" and "Estar" are used to express "To Be" in Spanish. Although they are use very differently. "Ser" is used when referring to: description, occupation, characteristic, time/date, origen, and relationship.
Examples:
Ella es una mujer = She is a woman.
Yo soy maestra de español. = I am Spanish teacher
Tú eres talentosa. = You are talented.
Today is Friday and it is 5:30 pm = Hoy es viernes y son las 5:30 pm.
Yo soy chilena = I'm Chilean.
Ella es mi amiga = She is my friend
On the other hand "Estar" is used when referring to: position, location, condition, emotion, and when using present continuous tense.
Examples:
Tú estás en el primer lugar. = You are in the first place.
Yo estoy en la escuela. = I'm in the school.
Ella está enferma. = She is sick.
Nosotros estamos felices hoy. = We are happy today.
Yo estoy jugando. = I'm playing.

The main differences between ser and estar include that ser is used for permanent qualities, like your name, your place of origin, and your physical appearance, while estar is used to talk about temporary situations, such as how you're feeling right now or your location.
