To design a PCR primer for the single-stranded DNA sequence you provided (5' - TACGGTAGGTC - 3'), we typically design primers that are complementary to the target sequence. For a primer that is 5 nucleotides long, we can choose a section from the sequence.
One option is to take the first 5 nucleotides from the 5' end:
5' - TACGG - 3'
However, if you want the primer to anneal to the complementary strand during PCR, the complementary sequence would be:
3' - AUGCC - 5'
But when writing it for PCR, we present it in the 5' to 3' direction:
5' - CCGTA - 3'
