Check your water that you are using for dilution as well as your phosphate buffer. Measure them on their own after blanking with a fresh stock of autoclaved distilled water. If you get a reading, most probably the water and/or buffer are contaminated. You replace them for your next set of experiments. Also have positive controls (known proteins) to see if the instrument is faulty. A bad UV bulb in the instrument will sometimes give inconsistent results.
