Is valediction in one line at the end of offical letter acceptable? For example: Sincerely yours, John Snow

I wouldn’t recommend using a valediction without the break after the comma. When you write it as one line, even in the context of a farewell, the recipient will instinctually read “Sincerely yours, John Snow” as a full sentence, if only for a split second.

Breaking after the comma is just good etiquette for official letters and emails.

Typically, no. English grammar would mean that your example could be interpreted to mean the reader is 'John Snow' and you the writer are claiming to be theirs; Sincerely YOURS, John Snow. The line break allows a space between the ideas and for your actual meaning to be clear.

Note: If there is an issue of space or characters, exceptions are always allowable.
