Assuming you're doing words to math and you're trying to translate your question into an equation, it'd be:
12-x
So, 12 minus "x" ("x" being the variable, a.k.a. "a number") We use the variable because we don't know what number "the number" is.
Difference (in math) means: the result of subtracting one umber from another. So how much one number differs from another.
That's how you know to take the 12 and subtract the "the number" (which is x)
