This question needs to be out in a context. Consider Ancient Mexico. The Aztecs conquered and enslaved other indigenous people, so they were enslaving locals long before European explorers arrived. If you want to know which European nation was first to enslave local indigenous peoples in the New World, I am reasonably sure it was Spain, though it did not take long for others to do the same.
