The bacterium that produces HindIII is Haemophilus influenzae, and this bug is usually found in the nasal passages and in the throat of humans. Is that what you mean?

That depends on how much is in the well. If you have 90 ul in the first well, add 10 ul of dye. And if each successive well has 90 ul, you can transfer 10 ul from the previous well to that one. But note that after transfer, the previous well will now have 90 ul but the final well will have 100 ul.
