Correct: Itʻs time for all of us to prepare for Mother's Day.
Itʻs = It is. This is a contraction for the subject it, and the verb is.
Motherʻs Day = ʻs denotes possessive singular.
However, grammatically speaking, it could be Mothersʻ Day (possessive plural) (a day to celebrate all mothers in the world) However, an interesting fact is that Motherʻs Day is the version used in the law that made the day an official holiday in the US, and so Motherʻs Day (singular possessive) is used as a precedent.
