from a 100mL prepared buffer, only 20mL was used for the addition of a strong acid or base. will the taken 20mL still represents the same moles as the originally prepared buffer solution?

Will the moles of the obtained 20mL buffer from the originally prepared 100mL buffered solution have the same moles or will they be different? What I am sure of is that the concentration of the 2 solutiom remains the same.
