When a gas enters an opened container and reacts with the substances in the container, is it part of the initial measurement of the reactants on the scale? Does the total mass change during a chemical reaction?

It's hard to say what the "initial measurement" is without more information. But if the reactants include the substances in the container as well as the gas, then yes I'd include the gas's mass for the measurement. Total mass doesn't change during a chemical reaction. Conservation of mass applies.
