The first "of" is a preposition to "half of that number."
The second "of" - in half of that number - is for multiplication.
The problem is very poorly written and IMHO worthless as an exercise.

but how you know that what you see in that sentence that leads to conclude that first of is preposition and other of is multiplication means .

I sense that English is not your native language.
I cannot provide a rule for determining if "of" is just a proposition or it also means multiplication.
Practice in use and hearing is my only suggestion.
